α-Lipoic Acid is a metabolic antioxidant that regulates NFκB signal transduction and can protect against oxidative injury. The exogenous compound is reduced intracellularly by enzymes. Its reduced form is involved in radical scavenging, recycling of other antioxidants, accelerating GSH synthesis, and regulating transcription factor activity, specifically NFkappaB. It also acts as a conenzyme that is required for the activity of pyruvate dehydrogenase and glycine decarboxylase, among other enzyme complexes. Studies show that it can prevent a decrease in the renal antioxidant defense system and prevent the increase of lipid peroxidation, platinum content and plasma creatinine concentrations. In other studies, lipoic acid decreased the phagocytosis of myelin by macrophages, acting as a non-specific scavenger of reactive oxygen species (ROS).
α-Lipoic acid and its reduced form, dihydrolipoic acid, are powerful antioxidants. α-Lipoic acid also scavenges hydroxyl radicals, hypochlorous acid, peroxynitrite, and singlet oxygen. Dihydrolipoic acid is a racemic compound but the R-enantiomer is thought to be more bioavailable compared to the S-enantiomer.
Details
Alpha-Lipoic Acid is a naturally occurring micronutrient, synthesized in small amounts by plants and animals (including humans), with antioxidant and potential chemopreventive activities. Alpha-lipoic acid acts as a free radical scavenger and assists in repairing oxidative damage and regenerates endogenous antioxidants, including vitamins C and E and glutathione. This agent also promotes glutathione synthesis. In addition, alpha-lipoic acid exerts metal chelating capacities and functions as a cofactor in various mitochondrial enzyme complexes involved in the decarboxylation of alpha-keto acids.
