Vonoprazan fumarate is a pyrole derivative and potassium-competitive acid blocker (P-CAB) that competitively blocks the potassium-binding site of gastric H (+),K (+)-ATPase, a key enzyme in the process of gastric acid secretion.

Vonoprazan is a selective, reversible, and potassium-competitive proton pump inhibitor that inhibits gastric H + /K + ATPase (IC 50 = 17 nM) but does not inhibit porcine Na + /K + ATPase activity when used at a concentration of 10 µM. 1, 2 It maintains its inhibitory effect in both weakly acidic (pH 6.5) and neutral (pH 7.5) conditions with IC 50 values of 19 and 28 nM, respectively.

What is Vonoprazan fumarat

Vonoprazan fumarate is a first-in-class potassium competitive acid blocker. 

It was approved in the Japanese market in February,2015. Vonoprazan can be used for the treatment of gastroduodenal ulcer, reflux esophagitis, and for some drug induced peptic ulcers. 

It can be combined with other antibiotics for the eradication of Helicobacter pylori.
